How to Reconstitute Lyophilized Peptides (3ml Vials)
Peptides are shipped as a lyophilized (freeze-dried) powder to preserve stability during transport. Before use, they must be reconstituted with a sterile diluent.
This guide outlines the standard process for 3ml peptide vials.
What You’ll Need
- Lyophilized peptide vial (3ml)
- Bacteriostatic water (preferred) or sterile water for injection
- Sterile syringe (1–3ml)
- Alcohol prep pads
- Clean, flat work surface

Step 1: Prepare a Clean Workspace
- Wash your hands thoroughly.
- Wipe down your work surface.
- Use alcohol pads to clean the rubber stoppers on both:
- The peptide vial
- The bacteriostatic water vial
Allow stoppers to air dry completely before proceeding.
Step 2: Draw the Diluent
Using a sterile syringe:
- Pull back the plunger to your desired amount of bacteriostatic water.
- Insert the needle into the water vial.
- Inject the air into the vial (this prevents vacuum resistance).
- Invert the vial and slowly draw out the desired amount of liquid.
Step 3: Add Diluent to the Peptide Vial
- Insert the needle into the peptide vial.
- Slowly inject the bacteriostatic water down the inside wall of the vial.
- Do not inject directly into the powder.
- Inject slowly to prevent foaming or damaging the peptide structure.
Step 4: Allow the Peptide to Dissolve
- Do not shake the vial.
- Gently swirl or roll the vial between your fingers if needed.
- Most peptides dissolve within a few minutes.
The solution should appear clear. If it is cloudy or contains visible particles, do not use.
Choosing How Much Water to Add
The amount of bacteriostatic water you add determines your final concentration.
Common dilution amounts for 3ml vials:
- 1ml total water → Higher concentration
- 2ml total water → Moderate concentration
- 3ml total water → Lower concentration (often easier for precise measurement)
Example
If you have a 5mg vial and add:
- 1ml → 5mg per ml
- 2ml → 2.5mg per ml
- 2.5ml → 2mg per ml
- 3ml → 1.67mg per ml
Choose a dilution that allows simple, accurate measurement for your intended protocol.
Storage After Reconstitution
- Store refrigerated (2–8°C / 36–46°F)
- Keep away from light
- Do not freeze
- When mixed with bacteriostatic water, use within approximately 30-60 days

- Always use sterile syringes.

- Avoid excessive agitation.
- Do not use if the solution changes color or develops particles.
